Research abstract
Fluorescent chromosome painting probes allow identification of a variety of chromosome abnormalitieinterphase and mitotic cells. Mouse cytogenetics, handicapped by difficult chromosome identificationchromosome paints in many research and commercial applications, including in vivo and in vitro genteWidespread commerical use of mouse painting probes requires reliability and ease of use in laboratorexperienced in mouse cytogenetics. Reliability depends on blocking probe hybridization to genomic reEase of use and versatility can be enhanced by simplifying fluorescence detection. This research proprobes using reliable mouse Cot1 repetitive DNA produced in house, increasing consistency of the finreducing pre-annealing time and cost. Alternatively, repeat sequences may be remove form painting prof repetitive sequences to immobilized repetitive DNA: resultant probe will be tested for painting.increase probe specificity and reduce the need for Cot1 pre-annealing. Labeling with different conjuallow users to combine probes and distinguish them by fluorescence. Direct labeling with fluorochromnucleotides may eliminate post-hybridization detection steps. Finally, the feasiblity of preparing frather than paint specific chromosomes for interphase chromosome counting and mapping will be examinbacterial artificail chromosome libraries for sequences mapping will be wexamined by screening bactechromosome libraries for sequences mapping to specific chromosome ends.
