Research abstract
Human cytochrome P450s play a prominent role in the metabolism of steroids, fatty acids, eicosanoids, drugs, phytochemicals and toxins. Specific enzymes can activate carcinogens, can beinduced, stimulated or inhibited by various compounds, and can display polymorphic forms. Althoughthe enzymic properties measured using animal P450s are suggestive reagents/systems are nonethelessimprecise predictors of the catalytic range and sensitivity associated with a particular human P450. Wewill address this deficiency. The long range goal is the development of suitable reagents and assaysystems that can readily evaluate high level heterologous expression systems that can readily evaluatehigh level heterologous expression systems as well as rapid and reliable site specific, high affinity, inhibitory antibodies will be evaluated. The availability of pure, cloned enzyme and specific antibodieswill help delineate the myriad possibilities for interaction between this complex enzyme family and a vastarray of pharmaceutical, environmental and dietary compounds. This knoledge can be essential forpredicting drug-durg interactions and the impact of genetic deficiencies on toxicity.
