Research abstract

Nitrone spin traps such as PBN, DMPO, and 4-POBN have made possible in vivo detection ofxenobiotic metabolites as radical adducts. Some of their deuterated and methylated analogs were foundor predicted to greatly improve the resolution and sensitivity of ESR or stability of the adducts, and mayallow further advances in this field. Phase I will develop the synthetic and purification proceduressuitable for commercial production of DMPO-d10, 4-POPBN-d13, and ¿-methylated analog of PBN.Syntheses of the B-methylated analogs of DMPO, 4-POBN, and some other promising spin traps maybe pursued in Phase II. The compounds will be verified to be free of ESR detectable impurities and usefulfor ESR spin-trapping study.
