Research abstract

THE FOUR SEROTYPES OF DENGUE VIRUSES ARE A LEADING CAUSE OF MORBIDITY THROUGHOUT THE TROPICS AND SUBTROPICS. FOR USE IN DIAGNOSTICS AND IN SUBUNIT VACCINES AGAINST DENGUE, A RELIABLE AND INEXPENSIVE SOURCE OF DENGUE ANTIGENS IS REQUIRED. FOR THESE PURPOSES, EXPRESSION OF RECOMBINANT ANTIGENS IN MICROORGANISMS IS BEST SUITED. WE PROPOSE TO USE THE YEAST SACCHAROMYCES CEREVISIAE TO PRODUCE THE DENGUE 2 VIRUS ENVELOPE GLYCOPROTEIN. THE SEQUENCES ENCODING THE ENVELOP PROTEIN WILL BE PLACED UNDER THE TRANSCRIPTIONAL CONTROL OF BOTH A CONSTITUTIVE AND A REGULATED PROMOTER, WITH AND WITHOUT SECRETION SIGNAL PEPTIDES. RELATIVE AMOUNTS OF ANTIGENIC MATERIAL WILL BE DETERMINED FOR EACH SYSTEM.
