Research abstract

Linoleic acid, the predominant polyunsaturated fatty acid (PUFA) in the human diet, can bemetabolized by cyclooxgenase, lipoxygenase and P450 enzymes. The monohydroxy derivatives oflinoleic acid, 9(S)-HODE and 13 (S)-HODE, are the most widely distributed of the known linoleic acidmetabolites. These compounds exhibit interesting biologic activities, including regulation of plateletfunction, maintenance of vascular thromboresistance and transduction of the cellular responses tocertain growth factors. The HODE derivatives of linoleic acid may also influence certain pathologicalstates including, psoriasis, the development of atherosclerosis and the development of cancer. Indeed, there is a strong positive correlation between the polyunsaturated fatty acid (PUFA) content of thehuman diet and the incidence of colorectal and breast cancers. Further studies on the physiologic andpathophysiologic roles of HODEs has been slowed by the lack of simple and convenient assays specificfor these compounds. Therefore, the ultimate goal of the present proposal is the development ofsensitive and specific immunoassays for these linoleic acid metabolites. Based on evidence obtained todate, it is also possible that the immunoassay reagents developed during the proposed studies will haveimportant diagnostic application in certain diseases, including atherosclerosis and cancer.
