Research abstract
Risk of transfusion-associated infection persists despite improved donor testing. Testing cannot prprospectively against new viruses which may enter the population, e.g., HIV. Solvent-detergent treatenveloped viruses, but cannot be carried out on single plasma units of fresh frozen plasma (FFP) inpathogens of importance for FFP transfusion include human immunodeficiency virus (HIV), hepatitis Bhepatitis C virus (HCV). The goal is to develop decontamination (PCD) methods, effective against at6) viruses per ml, to treat single FFP units in blood banks using S-59, a novel psoralen. This reseaaims: 1) demonstration of inactivation of HIV in plasma; 2) demonstration of inactivation of duckin plasma using DHBV as a model for HBV; 3) demonstration of inactivation of the flavivirus bovine v(BVDV) in plasma. BVDV is a model for HCV. 4) Dvaluation of the effects of PCD on coagulation protethe following in vitro assays: PT, a PTT, thrombin time, fibrinogen, Factor V, Factor VII, Factor IXFactor. 5) Evaluation of the mutagenicity of S-59 using the Ames' assay with and without S-59 activamicronucleus assay.
