Research abstract

In the proposed project the applicant will explore the development of a miniature MALDI-TOF mass spectrometer which combines for the first time the technologies of continuous flow (CF) MALDI with delayed ion extraction. The phase I studies will determine the compatibility of Bruker's unique CF interface and a recent Bruker invention for further improvements in delayed ion extraction. Experiments will be performed modifying existing instrumentation to evaluate the effects of both pressure and vacuum chamber size on the achievable mass resolution. From this data we will determine the minimum flight path and pumping required for a mass resolution and accuracy given by typical biomarkers used for triggering and identification of biological agents, while adhering to the goal of a compact instrument. This information will provide a basis for a concept design of a miniature continuous flow MALDI-TOF instrument. Features of the final design such as optimum operating voltages, minimal size and operating parameters will be established from phase I measurements. In the phase II portion of this project, the concept design developed in phase I will be used to construct a prototype instrument for testing, and evaluation of trigger/identification of simulants for biological agents. To commercialize an ultra-compact, low-cost benchtop MALDI-TOF instrument capable of providing automated analyses for routine analytical and diagnostic applications in research, quality-control and clinical-type laboratories.
