Research abstract

We propose to develop a sensitive, rapid, and affordable CRISPR-based diagnostic test for human parasites. The assay will be simple to run, require no instrumentation or advanced training, and be suitable for resourcelimited regions. In the Phase I, we will design and optimize the diagnostic for leishmaniasis using our multichambered FAST device. In a Phase II, additional development of the leishmania diagnostic or expansion to other parasites of concern may be proposed. In Objective 1, we will evaluate and optimize oligos for isothermal amplification, oligos for CRISPR sensing of target DNA, and selection of the readout using plasmidbased DNA targets. In Objective 2, we will collect and propagate a panel of leishmania species (and trypanosomes for specificity controls) and determine the specificity of the diagnostic. In Objective 3, we will focus on future manufacturing and scale-up of the FAST-CRISPR diagnostic. In Objective 4, we will develop IP and regulatory approval roadmaps.
