Research abstract

Recent advances in ultrafast, solid-state, near-infrared laser sources have led to the developmentlaser-based techniques in scientific instrumentation, including demonstrations of high-resolution, tof biophysical specimens using two photonexcitation fluorescence (TPF) laser scanning microscopy. Thcombines the spatial imaging properties of the laser scanning microscope (LSM) with TPF from naturalfluorphores, such as nicotinamide adenine dinucleotide (NADH), or staining dyes to study specimens aAn ultrafast laser source intended for TPF laser scanning microscopy in biophysics and biology shoul670 nm to 780 nm, provide sub-100 fs duration pulses at 100 MHz repetition rates, and have an averagKey development goals should be low cost (both capital and operating), small size, reliability, effioperation. In this effort we will address these goals with the development of a frequency-doubled, slaser. TPF laser scanning microscopy will be demonstrated using this source, with emphasis on excitaNADH at 680 nm with sub-micron spatial resolution.
