Research abstract

ERYTHROPOIETIN IS AN IMPORTANT HORMONE THAT REGULATES THE PRODUCTION OF RED BLOOD CELLS. IT IS PRODUCED IN THE KIDNEYIN RESPONSE TO HYPOXIA. SEVERAL ANEMIAS AND CYTHEMIAS ARE ASSOCIATED WITH ABNORMAL LEVELS OF ERYTHROPOIETIN. THE GOALOF THIS PROJECT IS TO DEVELOP A HIGHLY SENSITIVE AND ACCURATE ERYTHROPOIETIN ASSAY FOR USE IN ERYTHROPOIETIN RESEARCH AND FOR THE DIAGNOSIS OF DISEASES. SUCH AN ASSAY IS CURRENTLY NOT AVAILABLE COMMERCIALLY. IN PHASE I, MONOCLONAL AND POLYCLONAL ANTIBODIES WILL BE PRODUCED AGAINST TWO DIFFERENT SYNTHETIC ERYTHROPOIETIN PEPTIDES. ONE PEPTIDE CONSISTS OF AN INTERNAL HYDROPHILIC REGION OF THE ERYTHROPOIETIN MOLECULE, AND THE OTHER PEPTIDE INCLUDES THE CARBOXY TERMINAL AMINO ACIDS. THESE ANTIBODIES WILL BE TESTED FOR THEIR SUITABILITY FOR USE IN DIAGNOSTIC TESTS. THE PRODUCTION OF HIGHLY SPECIFIC ANTIBODIES AGAINST TWO POTENTIALLY DIFFERENT ERYTHROPOIETIN EPITOPES IN PHASE I WILL GREATLY FACILITATE THE DESIGN OF A NOVEL, EASY FORMAT FOR A HIGHLY ACCURATE AND SENSITIVE NONRADIOACTIVE ERYTHROPOIETIN IMMUNOASSAY IN PHASE II.
