Research abstract

There is currently no presymptomatic clinical diagnostic tool for the diagnosis of Parkinson's disedepletion of the dopamine transporter (DAT) in the striatum is a well-established marker of Pd and cdiagnosis of Pd can be achieved by a measure of DAT depletion. We will design and synthesize a technradiolabeled Single Photon Emission Computed Tomography ligand with high binding affinity and selectto be used in the measurement of DAT depletion and diagnosis of Pd. The proposal is based upon the dtropane analogs bind potently and selectively to the DAT and three tropane skeletons have been selecdifluoropine, 0-401, and WIN 35, 428. The novel ligands to be prepared will be composed of a tropane99mTc chelating unit. Three ligands have been selected for synthesis and biological evaluation in reduring Phase l. In Phase l a lead SPECT ligand will have been identified. If successful, a Phase IIsubmitted to optimize this lead. SPECT studies and detailed in vitro and in vivo pharmacology will b
