Research abstract

Project Summary Enabling high accuracy nanopore based strand sequencing has the potential to improve the understanding of long range genomic relationships and processes andbased on its accessibilityenable the associated clinical and diagnostic applicationsthe fastest growing sequencing sectorUnfortunatelyit is commonplace for nanopore sequencing error rates to exceedand higher accuracy is needed to advance nanopore sequencing applications toward assay specific applicationsAmong the various DNA sequencing technologiesnanopore strand sequencing has emerged as a very promising method to enable genomicstranscriptomicsepigenomicsand epitranscriptomics due to the currently associated advantagessuch as adaptabilitycostdirect read capabilitiesread lengthetcrelative to presently market dominating sequencing by synthesis technologiesHoweverthe library preparation procedures associated with nanopore based strand sequencing approachesalong with the resulting read accuracymust be improved before it will be widely adopted and standardized technology in the fieldThusto address the improvements needed to enable high accuracy nanopore based strand sequencingEBS proposes to develop broadly applicable methodology that will vastly improve both library preparation processes and the ability to control strand translocationwhich will provide significantly higher quality sequencing accuracy such that it will yield a nanopore based technology that can be confidently used for clinical or POC use Project Narrative Developing tools that enable improved genomic sequencingvia both both high accuracy and long read capabilitieshas the potential to advance clinical care via the associated diagnostic developmentsSuch achievements will enable routine genomic and precision medicine for comprehensivepersonalized patient screeningwhich in turn will lead to substantial improvements in population healthcare and survival
