Research abstract
Project Abstract
Automated sectioningimagingand extraction of renal glomeruli for single cell analysisUnderstanding heterogeneous cell states is essential for studying and treating diverse
diseases of the kidneyRenal glomerulim acrossare abundant but sparseand
implicated in many causes of chronic kidney diseaseThe proposed method will automate the
sectioningimagingand extraction of renal glomeruli with single cell resolutionThis approach
will sample large populations of glomeruli from human tissue and register serial sections into
digitald reconstructionsUsing this techniquethis project aims toquantify single cells in
whole glomeruliidentify a panel of compatible immunostaining markersand assess the
feasibility of capturing serial frozen tissue sectionsTogetherthese will validate new methods
to study cell distribution and protein expression in glomerular diseaseMore broadlythis will
facilitate additional automation of sectioningstainingimagingand analysis for single cell
experimentsThis will enable quantitativesingle cell research in renal pathologybenefiting
scientific discovery as well as the development of new treatments Project Narrative
Automated sectioningimagingand extraction of renal glomeruli for single cell analysisOne in seven Americans suffers from chronic kidney diseaseCKDa leading cause of
death and morbidity with limited options for early detectionThis project will develop a new
approach to study the three dimensional distribution of single cells in renal glomerulimicroscopic structures that are currently difficult to studyThis will help researchers better
understand causes of CKDpotentially leading to new treatments and earlier intervention
