Research abstract

Sand flies can cause illnesses such as Leishmaniasis caused by a parasite or sand fly fever due to sand fly fever viruses. The objective of this study is to develop a robust, field grade, screening assay for the detection of both leishmania parasites and/or sand fly fever viruses directly in sand flies. The test will utilize a multi-line lateral flow (dipstick) chromatography incorporated into a cassette that enables the simultaneous screening of both infectious agents. The proposed assay would be very a useful screening test to assess sand fly infestations for leishmania parasites and sand fly fever viruses. This would facilitate the treatment of military personnel presenting with febrile illness. It would also help in eradication programs. In this Phase II we will complete the definition of the target antigens for sand fly fever virus detection, develop specific antibodies and incorporate in the combined assay to detect both leishmania parasites and sand fly fever viruses in sand flies. The assay for leishmania is already at an advanced stage of development. The combined assay will be optimized and then evaluated in a sand fly leishmania infection model and subsequently in field testing to determine the sensitivity and specificity of the multiplex assay.
