Research abstract

The proposed project aims to develop a novel hydrogel-based drug delivery system for the treatment of traumatic brain injury (TBI), a significant health issue affecting both military personnel and civilians. Our innovative approach utilizes a bioinspired catechol-based chitosan hydrogel, which is biodegradable, highly malleable, and easily transportable. This hydrogel is designed to deliver multiple neuroprotective drugs directly to the injury site. Key features include excellent biocompatibility, rapid gelation, and strong adhesion to wet tissues, making it ideal for use in battlefield and other austere environments where traditional medical resources are limited. Our bioinspired chitosan hydrogel incorporates catechol functional groups to enhance adhesion to wet tissues, ensuring it remains securely in place to provide an effective barrier and support for therapeutic agents. The chitosan component offers inherent antimicrobial and hemostatic properties, enhancing the hydrogel's suitability for emergency medical applications. Our proprietary syringe system facilitates the formation of the hydrogel at the point of injury. This system allows adjustment of the hydrogel's mechanical properties, ensuring effective wound sealing, hemostatic and antimicrobial actions, and controlled drug release. The hydrogel will be evaluated for its mechanical properties, hemostatic activity, biocompatibility, and drug release kinetics using in vitro models in Phase I.This project builds on our prior work and strong expertise in hydrogel technologies, drug delivery systems, and biomedical engineering. Our experienced team, comprising experts in bioengineering, traumatic neurosurgery, and neurotoxicology, is well-equipped to advance this technology from concept to practical application. Previous studies conducted by our team have demonstrated the efficacy of similar hydrogel systems in wound healing and drug delivery applications, providing a solid foundation for this project.By addressing the current gap in effective, field-capable neuroprotective treatments for TBI, this project has the potential to revolutionize TBI care in both military and civilian settings. The goal is to enhance survival rates and improve outcomes for TBI patients by providing immediate and effective treatment at the point of injury. Our multidisciplinary approach ensures a comprehensive evaluation of the hydrogel system, incorporating state-of-the-art techniques in material science, pharmacology, and clinical practice to optimize its performance and safety. In conclusion, developing this hydrogel-based drug delivery system represents a significant advancement in TBI treatment, offering a practical, effective, and easily deployable solution to a critical medical challenge. The successful implementation of this technology will enhance immediate TBI patient care and contribute to long-term recovery and rehabilitation, improving the quality of life for those affected by this debilitating condition.
