Research abstract

Sciperio, Inc. proposes the development of the Organoid Research and Characterization for Live Electrophysiology (ORACLE) platform—an automated, sensor-integrated system designed to revolutionize organoid-based research for trauma and disease modeling. Traditional approaches to studying traumatic injuries, particularly traumatic brain injuries (TBI), rely on animal models that fail to accurately replicate human physiological responses. Human-derived induced pluripotent stem cell (iPSC) organoids offer a promising alternative, enabling scalable, reproducible models of soft tissue injury. However, current organoid generation methods are labor-intensive, costly, and lack real-time physiological characterization. The ORACLE platform addresses these challenges by integrating high-throughput organoid expansion, real-time morphological and biochemical monitoring, and automated electrophysiological interrogation using a microfluidic coculture system with embedded 3D microelectrode arrays (MEAs). This system will be guided by an AI-driven Director module that dynamically adjusts growth conditions, ensuring high-quality, physiologically relevant organoid models. ORACLE will be validated in collaboration with the Lybrand Lab at Texas Woman’s University, where neural organoids will be cultured and assessed for viability in modeling TBI and neurotrauma responses. In Phase I, Sciperio will develop schematics for the ORACLE platform, fabricate a microfluidic multi-organ coculture device, and establish protocols for high-throughput iPSC-derived organoid expansion. Additionally, the feasibility of additively manufactured 3D MEAs for real-time electrophysiological analysis will be explored. This integrated approach will provide a scalable, fully autonomous platform for trauma research, drug screening, and personalized medicine applications, significantly advancing the field of organoid-based biomedical research.
